Acremonium chrysogenum, abbreviated as A. chrysogenum, is an aerobic filamentous fungus with a complex cell wall structure and a specific life cycle. A. chrysogenum is a major strain for the industrial production of a β-lactam antibiotic, cephalosporin C (CPC). Although few filamentous fungi are used in the industrial production of antibiotics, the production of cephalosporins by submerged fermentation using A. chrysogenum is still the main choice. However, cephalosporin production by A. chrysogenum suffers from low yields and titers compared to penicillin production by Penicillium chrysogenum, both penicillin and cephalosporin are β-lactam antibiotics.

Given the limitations of wild A. chrysogenum strains, genetic engineering modification and metabolic engineering regulation of A. chrysogenum are important approaches to manipulate A. chrysogenum to obtain high-yielding mutant strains. two gene clusters on the chromosome of A. chrysogenum are involved in the biosynthesis of CPC. These gene clusters have been identified and intensively studied and they consist of pcbAB-pcbC, cefD1-cefD2, and cefEF and cefG, which encode enzymes that play different roles involved in CPC biosynthesis, respectively.
